"Cane-sugar Invert Analysis By HPLC Utilizing A Post-column Derivatization Reaction"

Abstract: An HPLC system based on a strong cation-exchange column (Bio-Rad Aminex Carbohydrate HPX-87), isocratic elution with water and refractometric detection showed poor sensitivity towards invert sugar, leading to poor precision and reproducibility. The sensitivity was improved by using a modification of the method of Wright and Van Niekerk (Food Chem., 1983, 10, 211; J. Agric. Food Chem., 1983, 31, 282), with a column of µBondapak NH2 and aqueous 80% acetonitrile as mobile phase, post-column derivatization with blue tetrazolium, and detection of the blue formazan with a model 441 Waters absorbance detector. Rectilinear responses (correlation coefficient 0.999 and 0.96 for laevulose and dextrose, respectively) were obtained for ~0.2 to 15 µg of sugars injected, and recoveries were 95 to 125% and 108 to 125%, respectively. Results on cane-sugar-refining samples are compared with those of the Lane and Eynon copper-reduction method.
