Abstract: Peanut sample preparation and extraction were carried out by AOAC methods 26.020 to 26.025 with samples blended with twice the sample weight of aqueous methanol in method 26.020 and with CHCl3 and CH2Cl2 - acetone (9:1) in place of benzene and CHCl3 - acetone (9:1), respectively, in method 26.022. Extracts were cleaned up on a mini-column of Florisil by a modification of AOAC method 26.019. Aflatoxins were determined by HPLC on a Nova-Pak phenyl cartridge (10 cm x 8 mm) equipped with a Spheri-5 MPLC guard cartridge (3 cm x 4.6 mm) and with aqueous 20% THF as mobile phase (1.5 mL min-1). Post-column derivatization was carried out with saturated aqueous iodine (0.4 mL min-1) in a reaction coil with fluorimetric detection at 418 nm (excitation at 365 nm). Detection limits were 0.1 ng g-1 of aflatoxins B1 and G1 and 0.06 ng g-1 of B2 and G2. Calibration graphs were rectilinear from 25 pg to 1.2 ng of B1 and G1 and 7.5 to 375 pg of B2 and G2. Recoveries and coefficient of variation are tabulated.
