Abstract: Stem and petiole tissue from transgenic potato plants containing β-glucuronidase gene was homogenized with 0.1 M phosphate buffer of pH 7, and glass beads were mixed with the product in the ratio 1:3. A column containing the modified beads was used at 25°C in a flow injection system for the determination of 4-methylumbelliferyl β-D-glucuronide, with 0.1 M phosphate buffer as mobile phase (2 mL/min) and fluorimetry of the liberated 4-methylumbelliferone. With operation at pH 7.5, the signal was linearly related to glucuronide concentration 25 mM; the detection limit was 1 µM. The corresponding non-transgenic potato tissue gave no response. The enzyme sensor retained 98% of its original response after two months but had a comparatively slow response time (12 min at 25°C).
