Abstract: The chromatograph and post-column reaction detector were as previously described (cf. Anal. Chem., 1986, 58, 2380). The mono-, di- and tri-glycerides, in acetone or THF solution, were separated by HPLC on a stainless-steel column (25 cm x 4 mm) of Hitachi gel 3057 (3 µm) operated at 30°C. The mobile phases were(I) ethanol - acetonitrile - water (10:12:3) and(II) ethanol - acetonitrile (13:7), at a flow rate of 0.8 mL min-1; initially, 100% of solvent(I) was used over 15 min and solvent(II) was increased by 4.67% min-1 to 70% and then by 1% min-1 to 100% during 30 min. Rectilinear response was obtained, e.g., for 1-monopalmitin in the range 0.2 to 121 nmol; the detection limit was 0.07 nmol. Results for standard mixtures, hydrogenated coconut oil - tallow oil mixture and a commercial emulsifier in a household product are shown.
