Abstract: Lymphocyte cells were lysed with ice-cold 10% HClO4, the extract was freeze-thawed in liquid N and left on ice to extract the inositol phosphates. The precipitated proteins were removed by centrifugation and nucleotides were removed with charcoal. The solution was analyzed by HPLC on two MonoQ columns (5 cm x 5 mm) with upward concave gradient elution with 0.02 mM HCl - 13.5 µM-YCl3 and 0.4 M HCl - 21 µM-YCl3. A solution consisting of 350 µM-2-(4-pyridylazo)resorcinol (I) and 1.6 M triethanolamine was used to allow online detection at 520 nm. Inositol tris-, tetrakis-, pentakis- and hexakis-phosphates were determined in the T-lymphocyte cells.
