Abstract: Placental chorionic plate, villus tissue and basal plate homogenates, and umbilical blood samples were deproteinized with acetonitrile. The supernatant solution, after centrifugation, was evaporated under N and the residue was dissolved in sodium diluent (0.27 M Na+; pH 2.2) for analysis on a Waters 840 HPLC Amino Acid System equipped with a column (25 cm x 4.6 mm) of sulfonated polystyrene - divinylbenzene copolymer (equilibrated with Na+). Elution was effected with a gradient of two Na+ buffers of pH 3.15 and 7.4 and post-column derivatization involved phthalaldehyde and 2-mercaptoethanol in borate buffer (pH 10.4). Fluorescence detection was at 410 nm (excitation at 340 nm). A Digital Pro-350 computer made necessary modifications for overlapping or clustered peaks. Analysis took 90 min per sample. Nine essential and six non-essential amino-acids were determined, with rectilinear response from 100 to 500 pmol, and the detection limit was as low as 50 pmol in some instances.
