Abstract: Total non-structural carbohydrates were extracted from shoots of Caucasian bluestem (Bothriochloa caucasica), rhizomes of johnsongrass (Sorghum halepense) and shoots and stem bases of winter wheat by hydrolysis with boiling 2.5 mM H2SO4 for 1 h (which also served to gelatinize starch). Following addition of 0.2 M acetate buffer and dilution with 0.5% of glucan 1,4-α-glucosidase, the samples (pH 4.4) were incubated at 50°C and centrifuged; aliquots of the supernatant solution were taken for flow injection analysis (details given) with 2.5% 4-hydroxybenzohydrazide(I) - 2 M NaOH or with K3Fe(CN)6(II). Nearly complete extraction of total non-structural carbohydrates from wheat and Caucasian bluestem occurred immediately but a 24-h incubation was necessary for johnsongrass. Calibration graphs for glucose and fructose were rectilinear by the method with use of I from 4.5 to 18.0 µg and from 4.5 to 22.5 µg, respectively, and by the method with use of II from 62 to 155 µg for either sugar. Advantages over the copper iodimetric technique are discussed.
