Abstract: The application of immunosensors in HPLC and FIA is discussed combining the selectivity of antibody binding with the sensitivity of a particular label (fluorophore, enzyme, radioactivity or chemiluminescence tag). The conditions of integrating the detectors into the chromatographic system are outlined: (i) matching the mobile phase with the detector; (ii) matching the detector response speed with the speed of separation. Detection limits in immunoassays and immunodetection (ID) formats are presented as functions of the residence time of the analyte on the column and the total column capacity. The issue is discussed of identifying more than a single antigenic species to the immobilized antibody ID cartridge. Evaluating and improving the recognition ability of the ID columns for detection of growth-hormone releasing factor is described, as well as problems of non-specific adsorption.
