Abstract: A new method for the direct determination of total fat in milk by use of a quartz crystal microbalance was developed. The method is based on a (micro) extraction procedure that is carried out in a flow injection system. Samples are diluted with an ethanol-water mixture and injected into the flow system, where they are mixed with n-hexane in an extraction coil. The analyte is determined in the organic phase. The calibration thus achieved is linear in the range 0.20-0.45% w/v, and the relative standard deviation is ± 3.2% (n = 11; P = 0.05). The throughput is 12 samples per hour. The proposed method was used to determine fat matter in milk samples; the results were found to be competitive with those of official methods for the same purpose.

Abstract: A review is presented of pre- and post-column derivatization methods for HPLC in which bifunctional compounds are used to react with 1,2-dioxo- or mono-oxo-compounds to yield fluorescent cyclic products. Examples are provided of applications to, e.g., aldehydes, 2-oxo-acids, catecholamines, peptides, fatty acids (including prostaglandins), nucleic acids derivatives and biogenic alcohols. Abstract: An ester solution (200 µL) was injected into an ethanol carrier stream (0.25 mL min-1) and merged with an ethanol stream containing 0.1 M tetrabutylammonium hydroxide (0.25 mL min-1). After mixing at 100°C in a 50-m reaction coil the stream was merged with 0.1 M butyric acid - ethanol containing 0.1 M LiCl and 0.2 µM-methyl red (0.5 mL min-1). The pH or absorbance of the mixed stream was measured at a glass electrode and/or at 490 nm. Calibration graphs were rectilinear from 0.02 to 0.1 M lauric acid ethyl ester; coefficient of variation was 1.3%. The sampling rate was ~20 samples h-1. The method was applied in the analysis of butter.

Abstract: Seventy-eight samples of [Scomberomorus brasiliensis (carite)] muscle tissue were analyzed for % fat, hypoxanthine content (determined. by a flow injection method), total volatile acids and bases, total bacterial count, and textural parameters (firmness, hardness, fracturability, cohesiveness, chewiness, and elasticity) were subjected to discriminant and factor analyzes. The apparent error rate obtained by discriminant anal. with all the parameters in the anal. was about 5.6%. However, classification was not improved (apparent error rate 6.9%) when cohesiveness and chewiness were excluded from the anal. The stepwise methods of selection (Wilks and Minresid) HxFI ranked second, and fourth when the Mahal and Maxminf methods were used. In the factor anal., using the principal component anal., five factors were extd. Factor loadings relate factor 1 (tastiness) with hypoxanthine concentration, total volatile acids and bases: factors 2, 3, 5 (texture) with the textural attributes; and factor 4 (rancidity) with the % fat content. Rotation of the reference axes showed that factors 1, 2 and 5 remained unchanged, while factors 3 and 4 were interchanged.

Abstract: The method is based on hydrolyis of neutral fat by triacylglycerol lipase (I) - carboxylesterase (II) and glycerol dehydrogenase (III)-catalyzed oxidation of glycerol, after which the NADH produced is monitored amperometrically. The cited sensor is made from I and II co-immobilized on aminopropyl-substituted controlled-pore glass and from III immobilized on Amino-Cellulofine. For application, an emulsion of milk in 6% bovine serum albumin solution is diluted with buffer solution prepared from 0.125 M carbonate buffer of pH 9.5 containing 30 mM (NH4)2SO4 and 1% of Triton X-100, and a portion is injected into the analyzer. (diagram given) for the determination of neutral fat at 25°C with the above-mentioned buffer as carrier stream at 1 mL min-1. When determining triolein, the relationship between response and concentration. is rectilinear for 0.0128 to 0.0853%; the coefficient of variation was 0.87%. Results were comparable with those obtained by the Wako triglyceride G-test method.
