Abstract: The method is based on the selective enzymatic cleavage of the cephalosporin β-lactam ring by highly pure penicillinase covalently bound to activated glass beads. Samples are injected into a phosphate buffer stream. The opening of the β-lactam ring causes significant decrease in u.v. absorbance. The difference between the absorbances at 254 nm of cephalosporin (measured after diversion of the stream through a dummy reactor tube containing only glass beads) and degraded cephalosporin is related rectilinearly to the concentration. of cephalosporin over the range 10 to 800 ppm (injection volume 0.1 ml). Results agreed well with those obtained by HPLC The coefficient of variation (n = 15) at the 207 ppm level was 0.7%, with a limit of detection of 2 ppm. The sample throughput is 30 h-1.

Abstract: Flow injection manifolds are described (with diagrams) for the determination of mono- and di-saccharides, lactate, amino-acids and glutamine in fermentation broths from the production of alkaline protease and penicillin. All manifolds contain immobilized enzyme cartidges and detection is by O-electrode. HPLC conditions for online and offline monitoring are also tabulated for determination of cephalosporins, penicillins, carbohydrates, amino-acids and inorganic anions.

Abstract: Sensitive and fast spectrophotometric methods for the determination of cephradine or ceftazidine in human urine, based on the formation of compounds between these drugs and Pd(II), are described. In the batch procedures the calibration graphs resulting from the measurement of the absorbance at 330 nm is linear over the range 5.0-60.0 µg mL-1 for cephradine and 3.0-60.0 µg mL-1 for ceftazidine. The methods were successfully adapted to FI-systems, the peak heights being proportional to cephalosporin concentration over the range 5.0-60.0 µg mL-1 for cephradine and 3.0-60.0 µg mL-1 for ceftazidine. The sampling frequency was 60 h-1 with a sample injection of 72 µL.

Abstract: Cephalosporin C(I) forms a reaction product with nicotinamide, which further forms a carbonyl addition product with dihydroxyacetone in alkaline medium. The UV absorbance of the final product is proportional to the concentration. of I. The potency of I in fermentation broths is determined by continuous-flow automatic analysis based on the above reactions; 30 samples can be analyzed in 1 h. The range of determination is up to 25,000 int. units mL-1, and results agree with those by HPLC. A flow diagram for the method is given. The method is simple, rapid, specific and sensitive.
